«ALSENSE Phytoplasma sp. RT-PCR» reagent kit
«ALSENSE Phytoplasma sp. RT-PCR» reagent kit for the detection of Phytoplasma sp. by RT-PCR method is designed for the detection of specific Phytoplasma sp. DNA by real time polymerase chain reaction.
Phytoplasmas are dangerous phytopathogens that cause disease in a wide range of plant species. They are a large group of cell wall-less bacteria belonging to the class Mollicutes and are obligate intracellular parasites of phloem tissues. Phytoplasma infections often lead to plant death and cause enormous damage to crop production worldwide. Some phytoplasmas (e.g. Candidatus Phytoplasma mali and Candidatus Phytoplasma pyri) are quarantine objects for the Republic of Belarus, the countries of the Eurasian Economic Union and countries that are members of the European and Mediterranean Plant Protection Organization.

Application field: The kit is suitable for research purposes.

«ALSENSE Phytoplasma sp. RT-PCR» reagent kit is based on a real time polymerase chain reaction (RT-PCR). The PCR method is based on the multiple doubling of a specific DNA region with DNA polymerase under artificial conditions (in vitro). As a result, DNA copies are produced in sufficient quantities to determine the result reliably. During the reaction, only a specific DNA region is copied that meets the conditions of specificity inherent in the structure of the primers and only if it is present in the sample under study.
The primers were designed based on the sequence of a conserved region of the 16S ribosomal RNA gene of several phytoplasma species, among which Candidatus Phytoplasma mali, Candidatus Phytoplasma pyri, Candidatus Phytoplasma prunorum, Candidatus Phytoplasma meliae and Candidatus Phytoplasma hispanicum are the most relevant. Detection of amplified DNA is provided by the intercalating fluorescent dye SybrGreen®.
Analytical specificity: 100%
Analytical sensitivity: To test the analytical sensitivity of the kit, a series of 8 ten-fold dilutions of Phytoplasma sp. DNA obtained from an infected plant with mild symptoms of bacterial blight was performed. A stable positive reaction was observed when the original sample was diluted 107 times.
