Specimens. Whole blood samples treated with such anticoagulants as citrate, EDTA, heparin are suitable for the study. Both fresh and frozen samples can be used. The yield and quality of purified DNA depends on the storage conditions.
Analytical characteristics. «ALPREP Spin Blood Manual» Reagent kit provides DNA extraction with purity (A260/A280 ratio) at least 1.8; the amount of DNA extracted is at least 2-10 mcg.

The extraction procedure involves proteinase sample treatment. Due to the presence of haotropic substances in lysis and precipitating buffers, DNA is adsorbed on the surface of the column silica-gel membrane, and then washed off possible inhibitors using washing buffers. A series of subsequent washes provides a highly purified DNA preparation, which is subsequently being dissolved in an eluting buffer and later is used in a PCR.
