«ALSENSE Phytoplasma pyri» reagent kit for the detection of Phytoplasma pyri by RT-PCR is based on a real-time polymerase chain reaction (RT-PCR) with hybridization-fluorescence detection on a DNA matrix. The PCR method is based on the multiple doubling of a specific DNA region with DNA polymerase under artificial conditions (in vitro). The amplification target is the 16S ribosomal RNA gene region of phytoplasmas of the 16SrX group. An internal control (ICS) was designed based on the 18S pear ribosomal RNA gene sequences.
The TaqMan probe to the Candidatus Phytoplasma pyri genome region carries a FAM dye tag at the 5′-end. The ICS-specific probe is labeled with the HEX fluorophore, allowing the user to validate the DNA extraction step from plants.
