«ALSENSE Phytoplasma mali/pyri» Reagent kit for the detection of Phytoplasma mali and Phytoplasma pyri by RT-PCR method is designed for the detection of specific Phytoplasma mali and Phytoplasma pyri DNA by PCR using TaqMan technology in nucleic acid preparations.
The apple tree proliferation phytoplasma (Candidatus Phytoplasma mali) and the pear decline and death phytoplasma (Candidatus Phytoplasma pyri) are the causative agents of dangerous diseases of apple and pear trees and cause serious economic damage to fruit growing in Europe. Phytopathogens belong to the taxonomic group 16SrX and are quarantine objects for the Republic of Belarus, the countries of the Eurasian Economic Union and countries that are members of the European and Mediterranean Plant Protection Organization (EPPO A2 list). The economically most important hosts for Candidatus Phytoplasma mali and Candidatus Phytoplasma pyri are Pyrus spp., Malus spp., Cydonia spp.

«ALSENSE Phytoplasma mali/pyri» reagent kit is based on the recommendations of the European and Mediterranean Plant Protection Organization (EPPO) and allows the detection of pathogen DNA by real time polymerase chain reaction. The target for amplification is the 16S ribosomal RNA gene site of phytoplasmas of the 16SrX group. An internal control (IC) was designed based on the 18S ribosomal RNA gene sequences of apple and pear trees.
The TaqMan® probe to the Candidatus Phytoplasma mali and Candidatus Phytoplasma pyri genome site carries a FAM dye label at the 5′-end. The ICS-specific probe is labeled with the HEX fluorophore, allowing the user to validate the DNA extraction step from plants.
Analytical specificity: 100%
Analytical sensitivity: To test the analytical sensitivity of the kit, a series of 8 ten-fold dilutions of Phytoplasma mali and Phytoplasma pyri DNA obtained from an infected plant with mild symptoms of bacterial blight was performed. A stable positive reaction was observed when the original sample was diluted 107 times.
