«ALSENSE Monkeypox virus kit» Real-Time PCR kit is designed to detect monkeypox virus DNA in clinical specimens by real-time polymerase chain reaction with hybridization-fluorescence detection in nucleic acid preparations isolated from clinical specimens (the recommended type of specimen for monkeypox laboratory confirmation is skin lesion material, including lesion and/or exudate swabs, covers from more than one lesion or lesion scab (crust)). The kit is intended for in vitro use only.

Application field: Research practice for the qualitative determination of monkeypox virus DNA.
User qualification requirements: The kit is intended for use by qualified personnel trained in molecular diagnostic techniques.
During PCR, a fragment of DNA is amplified multiple times in the presence of monkeypox pathogen genetic material. The detection of amplification products is performed during the analysis. For this purpose, special fluorescent labels (probes) are added to the reaction mixture, along with primers and other reaction components. The DNA probe carries a fluorescent label and a fluorescence quencher. When a specific product is formed, the DNA probe is destroyed and the quenching agent stops acting on the fluorescent label, resulting in an increase in the fluorescence level, which is detected by the amplifier. The reaction mixture includes oligonucleotides and probes for simultaneous amplification and detection of the DNA sections of the virus genome and the RNase P gene of the human genomic DNA (endogenous internal control).
The use of endogenous internal control makes it possible to:
- evaluate the correctness of biomaterial collection, transportation and storage;
- control the efficiency of nucleic acid isolation and PCR study stages, absence of reaction inhibition.
Amplification results are recorded using the following fluorescence detection channels (Table 1):
