Specimens. Whole blood samples treated with such anticoagulants as citrate, EDTA, heparin are suitable for the study. Both fresh and frozen samples can be used. The yield and quality of purified DNA depends on the storage conditions.
Analytical characteristics. «ALPREP M Sorb Blood» Reagent kit provides DNA extraction with a purity of not less than 1.7 according to the A260/A280 ratio; the recommended volume for DNA extraction is 200 µl.
The extraction procedure involves proteinase sample treatment. Due to the presence of chaotropic substances in the lysis and precipitation buffers, which improve lysis, the DNA is adsorbed on the surface of magnetic particles and then washed of possible inhibitors using wash buffers. A series of subsequent washes produces a highly purified DNA preparation that is subsequently dissolved in elution buffer and further used in PCR.

Part 1:
- Lysis buffer – 10 ml × 1 vial;
- Precipitating buffer concentrate – 10,5 ml × 1 vial;
- Washing buffer concentrate – 24 ml × 1 vial;
- Elution buffer – 5 ml × 1 vial;
- Magnetic sorbent – 1,5 ml × 1 test tube.
Part 2:
- Proteinase K (20 mg/ml) – 1 ml × 1 test tube.
