Description
PURPOSE
«ALPREP Forensic Q» Extraction kit is designed for manual DNA extraction or automated DNA extraction on the QIAcube Connect System (QIAGEN Germany) from forensic samples using spin-column membrane sorption. It is suited for DNA purification from trace samples (e.g., from crime scene); genotyping, including fingerprinting and paternity analysis; and routine analysis of reference samples. Suitable for research purposes, enabling the processing of human and animal biological samples for subsequent analysis via polymerase chain reaction and sequencing. «ALPREP BTA» Lysis Buffer is additionally used for DNA extraction from complex matrices such as dental and bone powder (not included in the kit).
ASSAY PRINCIPLE
The method is based on the reversible binding of DNA to the surface of silica gel column membrane. This is preceded by lysis and proteinase treatment of the sample. After sorption on the column membrane, DNA is washed from possible inhibitors using washing solutions, and then eluted in a buffer. DNA extraction from biological samples using the «ALPREP Forensic Q» Extraction kit can also be automated on the QIAcube Connect System (QIAGEN, Germany).
BIOMATERIAL FOR ANALYSIS
Tissue Material:
- soft tissues, including muscle tissue and dense fibrous tissue;
- soft tissues fixed in formalin and embedded in paraffin;
- nail material (nail plates and subungual content);
- hair fragments with hair follicles.
Dry and wet biological material traces:
- blood (whole blood, plasma, serum);
- seminal fluid (epithelial and sperm fractions after differential lysis);
- epithelium (buccal, skin);
- salivary secretions;
- sputum;
- urine;
- cerebrospinal fluid;
- bronchoalveolar lavage.
Biological material traces collected on carriers (paper, cotton swabs, FTA cards, etc.);
Adhesive materials (adhesive tapes, postage stamps, envelope flaps, cigarette butts, chewing gum, synthetic and natural fabrics, fabrics with complex fibrous structures, soil particles);
Complex matrices (in combination with «ALPREP BTA» lysis buffer):
- bone tissue samples;
- teeth.
