Description
PURPOSE
«ALPREP Plant-M Auto» Reagent kit is designed for DNA/RNA extraction from plant tissue samples (leaves, stems, roots, fruits, buds). It allows for nucleic acids extraction from frozen and dried samples as well. It can be used in research laboratories and by laboratories involved in phytosanitary control measures, species/variety identification of plants, and the detection of transgenic lines.
ASSAY PRINCIPLE
The principle of reagent kit involves the reversible binding of nucleic acids to the surface of magnetic particles, which can be easily precipitated from suspension using a magnetic stand.
The protocol for DNA/RNA extraction and purification includes:
- Sample preparation (homogenization of plant material);
- Lysis and precipitation of impurities with specialized buffers;
- Sorption of nucleic acids onto the magnetic sorbent and its subsequent precipitation on a magnetic stand;
- Two-stage washing of the sample;
- Elution and precipitation of magnetic particles followed by the collection of the supernatant containing nucleic acids.
The yield of nucleic acids depends on the plant species, storage conditions and duration, as well as the quality of homogenization.
