Description
PURPOSE
«ALPREP Milk-M Auto» Reagent kit is designed to automated extract DNA/RNA from milk samples (whole, pasteurized) and dairy products. Extracted DNA/RNA can be used for further analysis using PCR and reverse transcription PCR.
ASSAY PRINCIPLE
The principle of reagent kit is based on reversible binding of nucleic acids to the surface of magnetic particles, which can be easily precipitated from suspension using a magnetic stand after treatment with the kit’s components.
The protocol for DNA extraction and purification includes:
- Sample Preparation: Preliminary treatment of samples (manual procedures vary depending on the type of sample).
- Lysis and Contaminant Precipitation: Using specific buffers for cell lysis and impurity precipitation, followed by treatment of the reaction mixture with Proteinase K.
- Washing: A two-step wash of the sample.
- Nucleic Acid Sorption: Binding of nucleic acids to the magnetic sorbent followed by its precipitation on a magnetic stand.
- Elution: Eluting and precipitating the magnetic particles, then collecting the supernatant containing the nucleic acids.
BIOMATERIAL FOR ANALYSIS
- milk (whole, pasteurized);
- dairy products (kefir, sour cream, cream, cottage cheese, yogurt, butter, ice cream, etc.).
