Description
PURPOSE
«ALPREP Forensic Auto» Extraction kit is designed for nucleic acids extraction from human and animal biological material using an automated extraction system. Used at the pre-analytical stage of biological sample analysis in forensic practice for genetic testing, including species/group identification, individual identity verification, genotyping, and establishing biological relationships; as well as for scientific research purposes involving biological samples. Designed for the simultaneous extraction of DNA and RNA from 52/26/13 samples (on AutoMate Express or equivalent) and 32/96 samples (on 96-well format stations: Auto Pure, Gene Pure Pro, Nexor). «ALPREP BTA» Lysis Buffer is additionally used for nucleic acids extraction from complex matrices such as dental and bone powder (not included in the kit).
ASSAY PRINCIPLE
The method of magnetic sorption is used for DNA and RNA extraction which involves the reversible binding of nucleic acids to the surface of magnetic particles. The initial stage includes lysis and proteinase treatment of the sample. Due to the presence of chaotropic substances in the sorption buffer, nucleic acids adsorb onto the surface of magnetic particles and are then washed free of potential inhibitors using wash solutions. A series of washes effectively separates the components of cell lysate, ensuring the production of highly purified nucleic acid preparations suitable for use in PCR and other molecular applications.
BIOMATERIAL FOR ANALYSIS
Tissue Material:
- soft tissues, including muscle tissue and dense fibrous tissue;
- soft tissues fixed in formalin and embedded in paraffin;
- nail material (nail plates and subungual content);
- hair fragments with hair follicles.
Dry and wet biological material traces:
- blood (whole blood, plasma, serum);
- seminal fluid (epithelial and sperm fractions after differential lysis);
- epithelium (buccal, skin);
- salivary secretions;
- sputum;
- urine;
- cerebrospinal fluid;
- bronchoalveolar lavage.
Biological material traces collected on carriers (paper, cotton swabs, FTA cards, etc.);
Adhesive materials (adhesive tapes, postage stamps, envelope flaps, cigarette butts, chewing gum, synthetic and natural fabrics, fabrics with complex fibrous structures, soil particles);
Complex matrices (in combination with «ALPREP BTA» lysis buffer):
- bone tissue samples;
- teeth.
