Description
PURPOSE
«ALPREP Gel Column» Purification kit is designed for DNA extraction from agarose gel after gel electrophoresis and purification using microcentrifuge columns with sorption membranes. The purified DNA is suitable for subsequent use in PCR, qPCR, restriction, ligation, sample preparation for Sanger and NGS sequencing, as well as in other applications of genetic engineering.
ASSAY PRINCIPLE
The principle of reagent kit involves the reversible binding of nucleic acid molecules to the surface of a silica membrane in a spin column after processing with the kit’s components.
The protocol for DNA extraction and purification includes:
- Treatment of the agarose gel slice with a specialized buffer;
- Incubation in the membrane column to bind the dissolved DNA;
- Centrifugation to remove gel components, washing the DNA bound to the membrane;
- Elution followed by centrifugation to collect the purified DNA.
