«ALCHEM MgCl2 50 mM» Solution is used for optimization of Mg2+ ion concentration in the PCR Master mix. A typical final concentration for Mg2+ in PCR is in the range of 1–4 mM, with 0.5 mM titration increments recommended for optimization.
Mg2+ ions:
- Ensure the polymerase activity;
- Are necessary for coordinating dNTPs in the enzyme’s active site, which facilitates the transfer of phosphate groups and the formation of phosphodiester bonds in the new DNA strand;
- Affect the hybridization temperature of primers with DNA;
- Provide specificity and efficiency of the PCR reaction.
