Description
PURPOSE
The level of IgG to various proteins (including RBD) of the SARS-CoV-2 virus does not always correlate with their ability to prevent the penetration of viral particles into cells and the development of coronavirus infection. Among all the antibodies produced as a result of an infection or in response to a vaccine, only a part has a neutralizing activity (i.e., protects the body from infection with SARS-CoV-2).
Neutralizing antibodies block the interaction of the RBD site of SARS-CoV-2 with the ACE2 receptor on the human cell surface and are crucial for the protection of the organism against coronavirus infection. Therefore, determination of the level of neutralizing antibodies is important both for testing the efficacy of immunization with all types of vaccines and for determining the strength of post-infection immunity, and allows the risk of re-infection to be assessed with a high degree of confidence.
«ALMUNO-nAb-SARS-CoV-2» Kit is designed for semi-quantitative determination of antibodies with virus neutralizing activity in human serum or blood plasma based on the assessment of inhibition of binding of RBD-part of SARS-CoV-2 virus to ACE2 receptor.
The kit is designed for 96 determinations, including staging in triplicate of the negative control and in monoplicate of the positive control. The kit is intended for in vitro use only.
PRINCIPLE OF METHOD
The detection is based on competitive ELISA analysis. During incubation, biotinylated ACE2 binds to the RBD immobilized in the microplate wells. Neutralizing antibodies inhibit the binding of ACE2 to the RBD if they are present in the test sample. The streptavidin-HRP conjugate detects ACE2-biotin bound. The optical density of the solution is measured after incubation with substrate and adding a stop reagent. The measured optical density is inversely related to the amount of neutralizing antibodies to the SARS CoV-2 virus in the control and test samples.
