Description
PURPOSE
«ALMUNO Penicillin» Kit is a test system based on a recombinant penicillin-binding protein designed by Algimed Techno.
The kit enables the quantitative determination of penicillin-group antibiotics using the competitive enzyme-linked immunosorbent assay (ELISA) method in the following matrices:
- Milk (raw, pasteurized, sterilized), cream, reconstituted dry milk, dairy desserts, condensed milk, milk ice cream, dairy-based drinks and cocktails (including yogurt-based), milk porridges, reconstituted dry milk porridges, infant formulas and reconstituted dry milk infant formula;
- Butter, milk fat, spread;
- Cheese, cheese products, cheese pastes, and culinary cheese-based products;
- Cottage cheese and cottage cheese products (including cottage cheese bars, granular cottage cheese, cottage cheese mass, cottage cheese paste, cottage cheese creams, and similar products with or without fillers), cottage cheese-based cheese, fermented dairy products (ayran, yogurt, acidophilin, and similar products with or without fillers), sour cream, buttermilk, whey, reconstituted dry whey, whey-based beverages, permeate;
- Meat, meat products, meat-vegetable products, lard, pork fat, animal fats, by-products;
- Poultry meat, poultry products, semi-finished poultry products, canned poultry products, poultry by-products;
- Fish, aquacultural products, including infant food (culinary fish products and semi-finished products, canned and preserved fish, caviar, boiled-frozen fish food products), shrimps;
- Eggs, egg products, and processed egg products (including egg powder, liquid egg mass, and melange);
- Honey, creamed honey;
- Blood serum and plasma.
PRINCIPLE OF METHOD
The method is based on a competitive colorimetric enzyme-linked immunosorbent assay (ELISA). During the analysis, a biotinylated penicillin-binding protein (PBP) is added to the wells of a microplate coated with the antibiotic, along with the sample. If the sample contains a penicillin-group antibiotic, it competes with the coated antibiotic for binding to the PBP, thereby preventing its attachment to the microplate wells. The resulting complexes are detected using a streptavidin-peroxidase conjugate. The mass concentration of the penicillin-group antibiotic, expressed as benzylpenicillin, is determined based on a calibration curve constructed with six penicillin G calibration solutions.
