PATHOGEN INFORMATION
Gram-negative phytopathogenic bacteria Acidovorax citrulli cause a dangerous disease – bacterial blotch of pumpkin crops. Such economically important plants as watermelon, melon, pumpkin, cucumber, courgette and patisson are susceptible to the pathogen. The pathogen is a quarantine organism for countries of the Eurasian Economic Union and countries belonging to the European and Mediterranean Plant Protection Organisation (EPPO A1 list).
ASSAY PRINCIPLE
«ALSENSE Acidovorax citrulli» Reagent kit has been developed based on recommendations of the European and Mediterranean Plant Protection Organization (EPPO). The reagent kit is a multiplex, real-time PCR-based assay. The target for amplification is the mcpQ gene region of Acidovorax citrulli. The internal control (IC) is designed based on the 18S ribosomal RNA gene sequence of host plants. The TaqMan® probe to a region of the Acidovorax citrulli genome is labeled with a FAM fluorophore dye on the 5′-end. The IC-specific probe is labelled with a HEX fluorophore, allowing the user to validate the DNA extraction step from plants.
SAMPLING RECOMMENDATIONS
«ALSENSE Acidovorax citrulli» is validated for the simultaneous detection of Acidovorax citrulli & plant DNA by real-time PCR. Samples for diagnosis should consist of affected fruits and leaves, seeds can be used as test object. Samples are collected during the period of active growth, which usually lasts from late spring to autumn.
