Description
PURPOSE
«CleanCheck DNAse-RNAse Free» Real-Time PCR kit is designed for the detection of nuclease contamination. It allows detection of the lowest amounts of RNase and DNAase degrading both single- and double-stranded DNA. It can be used for inspection of plastic products and for detection of nuclease contamination in PCR laboratories on all types of surfaces, solutions, reagents (not PCR inhibitors), plastic consumables, etc.
PRINCIPLE OF METHOD
The principle of the method is based on the incubation of the sample (flush) in sample buffer with subsequent amplification of nucleic acids by polymerase chain reaction with hybridisation-fluorescence detection in the “real time” mode. To collect a sample from solid surfaces, a probe-tampon is used to wash off the sample. When controlling reagents or liquid solutions for the presence of contamination, the probe-tampon is placed in the solution under study. Selected samples are incubated for 2 hours at room temperature; at the end of incubation, real-time RT-PCR is performed.
