Description
PURPOSE
The ability to classify human white cells based on their expression of cell surface antigens using flow cytometry provides valuable diagnostic and prognostic information in a variety of pathologies. One of the steps in sample preparation is the removal of red blood cells from the whole blood sample prior to analysis of white blood cells on a flow cytometer.
«ALFLOW Prep 10X» kit consists of 10-x concentrate of lysing reagent based on ammonium chloride and a fixative solution. The reagent kit is intended for removal of erythrocytes from whole blood sample during preparation of human blood samples for analysis by flow cytometry.
PRINCIPLE OF THE PROCEDURE
The principle of the procedure is based on the ability of lysing reagent to destroy erythrocytes selectively, leaving other cells intact. The lysing reagent is added to a human whole blood sample during sample preparation stage. After incubation with the lysing reagent, which has a targeted effect, the erythrocyte membrane is destroyed into fragments. As a result, the red blood cell signal does not affect the accuracy and reliability of the analysis of target cell populations such as leukocytes. The addition of fixative solution stabilizes the leukocyte membranes, which increases the storage time of the prepared sample until the measurement is performed.
