Description
PURPOSE
The ability to classify human white cells based on their expression of cell surface antigens using flow cytometry provides valuable diagnostic and prognostic information in a variety of pathologies. One of the steps in sample preparation is the removal of red blood cells from the whole blood sample prior to analysis of white blood cells on a flow cytometer.
«ALFLOW AutoPrep» Lysing Reagent kit is designed for fast lysis of erythrocytes with simultaneous fixation of leukocytes, without washing step, when preparing human whole blood samples for analysis by flow cytometry. The reagent kit is used for manual or automatic lysis of erythrocytes using automated sample preparation stations*.

PRINCIPLES OF THE PROCEDURE
The principle of the procedure is based on the ability of lysing reagent to destroy erythrocytes selectively, leaving other cells intact. During sample preparation, reagents are sequentially added to a human whole blood sample. Reagent A lyses red blood cells, reagent B restores the sample pH and stabilizes white blood cells, and reagent C fixes the white blood cell membrane. As a result, the red blood cell signal does not affect the accuracy and reliability of the analysis of target cell populations such as leukocytes during analysis on a flow cytometer.
