The kit is intended for the qualitative determination of the content of class G immunoglobulins to SARS-CoV‑2 in human serum or plasma by enzyme immunoassay. The kit is intended for in vitro use only.

SARS-CoV‑2 is an enveloped single stranded (+) RNA virus belonging to the genus Betacoronavirus. First identified at the end of 2019. After entering the human body, it can cause a dangerous infectious disease – COVID-19, which can occur both in the form of an acute respiratory viral infection of the mild course, and in a severe form, the specific complications of which may include viral pneumonia, which entails acute respiratory distress syndrome or respiratory failure with risk of death.

Immunoglobulins G (IgG) are a class of antibodies that make up about 75% of human plasma antibodies and are the most commonly found antibodies in the bloodstream. IgGs take part in controlling infection throughout the body by binding to a wide variety of pathogens: viruses, bacteria, fungi. Binding of immunoglobulins G to pathogens causes them to immobilize and bind to each other (agglutination). IgG is involved mainly in the secondary immune response, is secreted as monomers that easily penetrate into tissues. Class G immunoglobulins are the only type of antibodies capable of crossing the placenta using special receptors, providing protection for the fetus in utero.

Determination of the content of IgG to SARS-CoV‑2 in blood plasma can help determine the formation of an immune response in patients with COVID-19 disease, as well as to assess the level of antibodies in the blood plasma of donors who have recovered.

The principle of operation of the kit is based on direct non-competitive ELISA analysis. The test and control samples are incubated in the wells of the plate coated with the recombinant nucleocapsid protein. If the sample contains immunoglobulins to this protein, they will specifically bind to the protein immobilized in the plate wells. At the next stage, antibodies of the IgG class are detected using the second antibodies in the peroxidase conjugate. After incubation with the substrate and the addition of the stop reagent, the optical density of the solution is measured at 450 nm. The measured optical density is in direct proportion to the amount of immunoglobulins G in the control and test samples.
